Description: Ig delta chain C region is an allelic product of the human IGHD gene. The two known IGHD alleles, IGHD*01 and IGHD*02, respectively produce isoforms 1, a secreted protein, and 2, a single-pass type I membrane protein. A member of the adaptive immune system, IgD are monomers expressed by activated B cells. Containing 3 Ig-like (immunoglobulin-like) domains, IgD chain C is located on chromosome 14 within the human heavy chain locus, lying on the 3? side of the IgM chain C region from the V-D-J cassette. Polyadenylation at certain sites along the heavy chain locus likely effects the mechanism that determines the alternative splicing event which results in the expression of either IgD chain C or IgM chain C. Some studies have suggested that antigenic coactivation of IgD+ B cells can have a negative influence on bone resorption during infectious events.

Description: Auxiliary protein of DNA polymerase delta and is involved in the control of eukaryotic DNA replication by increasing the polymerase's processibility during elongation of the leading strand. Induces a robust stimulatory effect on the 3'-5' exonuclease and 3'-phosphodiesterase, but not apurinic-apyrimidinic (AP) endonuclease, APEX2 activities. Has to be loaded onto DNA in order to be able to stimulate APEX2. Plays a key role in DNA damage response (DDR) by being conveniently positioned at the replication fork to coordinate DNA replication with DNA repair and DNA damage tolerance pathways. Acts as a loading platform to recruit DDR proteins that allow completion of DNA replication after DNA damage and promote postreplication repair: Monoubiquitinated PCNA leads to recruitment of translesion (TLS) polymerases, while 'Lys-63'-linked polyubiquitination of PCNA is involved in error-free pathway and employs recombination mechanisms to synthesize across the lesion.

Description: E3 ubiquitin ligase that, together with UBE2N and UBE2V1, mediates the synthesis of 'Lys-63'-linked-polyubiquitin chains conjugated to proteins, such as IKBKG, IRAK1, AKT1 and AKT2. Also mediates ubiquitination of free/unanchored polyubiquitin chain that leads to MAP3K7 activation (By similarity). Leads to the activation of NF-kappa-B and JUN. May be essential for the formation of functional osteoclasts. Seems to also play a role in dendritic cells (DCs) maturation and/or activation. Represses c-Myb-mediated transactivation, in B-lymphocytes. Adapter protein that seems to play a role in signal transduction initiated via TNF receptor, IL-1 receptor and IL-17 receptor. Regulates osteoclast differentiation by mediating the activation of adapter protein complex 1 (AP-1) and NF-kappa-B, in response to RANK-L stimulation. Together with MAP3K8, mediates CD40 signals that activate ERK in B-cells and macrophages, and thus may play a role in the regulation of immunoglobulin production.

Description: GPRC6A (G protein-coupled receptor family C group 6 member A) is a 928 amino acid protein encoded by the mouse Gprc6a gene. GPRC6A is a member of the G protein-coupled receptor 3 family. This receptor is activated by both amino acids and extracellular concentration of calcium ions. The activity of GPRC6A is mediated by a G protein that activates a phosphatidylinositol-calcium second messenger system. GPRC6A senses changes in the extracellular concentration of calcium ions, suggesting that it may mediate extracellular calcium-sensing responses in osteoblasts. Osteocalin stimulates the activity of GPRC6A in the presence of calcium. GPRC6A also acts as a receptor for amino acids, with a preference for basic amino acids such as L-Lys, L-Arg and L-ornithine. Its affinity for amino acids suggests that it may act as a regulatory component of the urea cycle. GPRC6A is expressed at high levels in liver, lung, spleen and heart, and at lower levels in kidney, skeletal muscle and brain.

Description: T2R16 is a 291 amino acid multi-pass membrane protein that belongs to the G-protein coupled receptor T2R family. Acting in the oral cavity and the gastrointestinal tract, T2R16 is a gustducin-coupled receptor that is implicated in the perception of bitter compounds. T2R16 mediates responses to certain taste through PLC ∫2, a phospholipase C selectively expressed in taste tissue, and the calcium-regulated cation channel TRPM5. Expressed in a subset of gustducin-positive taste receptor cells of the tongue and epithelia, T2R16 confers bitter perception of salicin to non-taster mice. The gene that encodes T2R16 consists of 996 bases and maps to human chromosome 7q31. Chromosome 7 houses over 1,000 genes, comprises nearly 5% of the human genome and has been linked to Osteogenesis imperfecta, Pendred syndrome, Lissencephaly, Citrullinemia and Shwachman-Diamond syndrome. The Lys-172 polymorphism in T2R16 is associated with genetic susceptibility to alcoholism.
