Description: The HUNK (hormonally upregulated Neu-associated kinase) protein, also designated MAK-V in mouse, has been identified as a novel SNF1-related serine/threonine kinase. The human HUNK gene localizes to chromosome 21q22 and encodes a protein with nucleocytoplasmic distribution and localizes to the centrosome. Overexpression of the HUNK protein associates with approximately 50% of breast carcinomas, and may provide diagnostic-prognostic value as a molecular marker. Serine/threonine-protein kinase SNF1-like kinase 2 (SIK) phosphorylates Ser-794 of IRS1 in insulin-stimulated adipocytes, which may modulate the efficiency of insulin signal transduction. SIK is activated by phosphorylation on Thr-175 by STK11 in complex with STE20-related adapter- and CAB39.

Description: A hallmark of signal transduction pathways is the reversible phosphorylation of serine and threonine residues within specific sequences, or motifs, in target proteins. Specific signaling motifs include not only sequences that are recognized by protein kinases, but also those that are recognized by phosphorylation-dependent binding proteins like 14-3-3. These modular phosphoprotein interacting domains are critical elements in modulating, directing and amplifying intracellular communications. Many critical protein kinases can be regulated by phosphorylation at a specific serine or threonine surrounded by phenylalanine or tyrosine. For example, Akt, an important kinase that regulates cell survival, is activated by phosphorylation at Ser473, a site surrounded by phenylalanine and tyrosine. RSK1, p70 S6 K, and certain PKC isoforms also contain a similar consensus phosphorylation site. Phosphorylation of these sites is required for kinase activity. The (Ser/Thr) Phe Antibody is a powerful tool for discovery of new proteins containing this important regulatory motif.

Description: Protein Kinase c alpha (PKC alpha) is an 77 kDa member of the conventional group (cPKCs: sensitive to calcium, diacylglycerol, phosphatidylserine and phorbol esters) of the PKC family of serine/ threonine kinases that are involved in a wide range of physiological processes including mitogenesis, cell survival and transcriptional regulation. PKC alpha is an ubiquitously expressed PKC isozyme that has been implicated in the regulation of a broad range of cellular functions including proliferation, differentiation, development, migration, cell cell adhesion, cell extracellular matrix adhesion, and solute transport. The activation loop threonine (threonine 497 in PKC alpha) of conventional PKCs is phosphorylated by phosphoinositide dependent kinase 1 (PDK1).
