Supplier: Bioss
Description: Serine-threonine kinase which transduces inflammatory and cell-death signals (programmed necrosis) following death receptors ligation, activation of pathogen recognition receptors (PRRs), and DNA damage. Upon activation of TNFR1 by the TNF-alpha family cytokines, TRADD and TRAF2 are recruited to the receptor. Phosphorylates DAB2IP at 'Ser-728' in a TNF-alpha-dependent manner, and thereby activates the MAP3K5-JNK apoptotic cascade. Ubiquitination by TRAF2 via 'Lys-63'-link chains acts as a critical enhancer of communication with downstream signal transducers in the mitogen-activated protein kinase pathway and the NF-kappa-B pathway, which in turn mediate downstream events including the activation of genes encoding inflammatory molecules. Polyubiquitinated protein binds to IKBKG/NEMO, the regulatory subunit of the IKK complex, a critical event for NF-kappa-B activation. Interaction with other cellular RHIM-containing adapters initiates gene activation and cell death. RIPK1 and RIPK3 association, in particular, forms a necrosis-inducing complex.

Description: F-box proteins are critical components of the SCF (Skp1-CUL-1-F-box protein) type E3 ubiquitin ligase complex and are involved in substrate recognition and protein recruitment. F-box proteins are members of a large family that regulate the cell cycle, immune response, signaling cascades and developmental programs by targeting proteins, such as cyclins, for degradation by the proteasome after ubiquitination. FBL11, also known as FBXL11 (F-box and leucine-rich repeat protein 11), CXXC8, KDM2A, JHDM1A (JmjC domain-containing histone demethylation protein 1A) or LILINA, is a 1,162 amino acid member of the F-box protein family that contains one F-box domain and localizes to the nucleus. Expressed ubiquitously with highest expression in testis, ovary and brain, FBL11 functions to demethylate the Lys-36 residue of histone H3, thereby modulating the histone code. Additionally, FBL11 is thought to promote the ubiquitination and subsequent degradation of various phosphorylated proteins. Three isoforms of FBL11 exist due to alternative splicing events.

Description: NAD-dependent protein deacetylase that specifically mediates deacetylation of histone H3 at 'Lys-18' (H3K18Ac). In contrast to other histone deacetylases, displays selectivity for a single histone mark, H3K18Ac, directly linked to control of gene expression. H3K18Ac is mainly present around the transcription start site of genes and has been linked to activation of nuclear hormone receptors. SIRT7 thereby acts as a transcription repressor. Moreover, H3K18 hypoacetylation has been reported as a marker of malignancy in various cancers and seems to maintain the transformed phenotype of cancer cells. These data suggest that SIRT7 may play a key role in oncogenic transformation by suppresses expression of tumor suppressor genes by locus-specific deacetylation of H3K18Ac at promoter regions. Also required to restore the transcription of ribosomal RNA (rRNA) at the exit from mitosis: promotes the association of RNA polymerase I with the rDNA promoter region and coding region. Stimulates transcription activity of the RNA polymerase I complex. May also deacetylate p53/TP53 and promotes cell survival, however such data need additional confirmation.

Description: The D7 monoclonal antibody specifically reacts with mouse Ly-6A/E (Sca-1), a 18 kDa GPI-linked surface protein. It is expressed on multipotent hematopoietic stem cells (HSC) and subsets of B and T cells and involved in B and T cell lymphocyte responses. Splenocytes from Ly-6.2 strains exhibit a larger number of D7-positive cells than those from the Ly-6.1 strains.
