Catalog Number: (BOSSBS-10024R-HRP)
Supplier: Bioss
Description: Responsible for the deacetylation of lysine residues on the N-terminal part of the core histones (H2A, H2B, H3 and H4), and some other non-histone substrates. Histone deacetylation gives a tag for epigenetic repression and plays an important role in transcriptional regulation, cell cycle progression and developmental events. Histone deacetylases act via the formation of large multiprotein complexes. Participates in the BCL6 transcriptional repressor activity by deacetylating the H3 'Lys-27' (H3K27) on enhancer elements, antagonizing EP300 acetyltransferase activity and repressing proximal gene expression. Probably participates in the regulation of transcription through its binding to the zinc-finger transcription factor YY1; increases YY1 repression activity. Required to repress transcription of the POU1F1 transcription factor. Acts as a molecular chaperone for shuttling phosphorylated NR2C1 to PML bodies for sumoylation.

Description: Testis-specific DNA binding protein responsible for insulator function, nuclear architecture and transcriptional control, which probably acts by recruiting epigenetic chromatin modifiers. Plays a key role in gene imprinting in male germline, by participating in the establishment of differential methylation at the IGF2/H19 imprinted control region (ICR). Directly binds the unmethylated H19 ICR and recruits the PRMT7 methyltransferase, leading to methylate histone H4 'Arg-3' to form H4R3sme2. This probably leads to recruit de novo DNA methyltransferases at these sites (By similarity). Seems to act as tumor suppressor. In association with DNMT1 and DNMT3B, involved in activation of BAG1 gene expression by binding to its promoter. Required for dimethylation of H3 lysine 4 (H3K4me2) of MYC and BRCA1 promoters.

Description: HDAC7 is a member of the class II mammalian histone deacetylases, which plays an important role in modulating the eukaryotic chromatin structure. Human HDAC7 is composed of 912 amino acid residues. Although HDAC7 is localized mostly to the cell nucleus, it is also found in the cytoplasm, suggesting nucleo-cytoplasmic shuttling. The histone deacetylase activity of HDAC7 maps to a carboxy-terminal domain and is dependent on interaction with class I HDACs in the nucleus. It is an active component of different transcriptional corepressor complexes that can be recruited to specific promoter regions via interactions with a growing number of sequence specific transcriptional factors. HDAC7 catalyzes removal of acetyl-groups from acetyl-lysines of histones and promotes compaction of chromatin in these regions, leading to the inhibition of gene transcription.

Description: ESET is a nuclear protein belonging to the histone-lysine methyltransferase family and to the Suvar3-9 subfamily. It is a highly conserved protein of 150 amino acids that has been implicated in chromatin structure modulation. ESET is excluded from cell nucleoli and areas of condensed chromatin and can associate with the nonpericentromeric regions of chromatin. The gene encoding for this protein, SETDB1, maps to chromosome 1q21. ESET is a histone methyltransferase, methylating Lys-9 of histone H3 and mutations within the SETDB1 gene abolishes its methyltransferase activity. This methylation acts as a tag for epigenetic transcriptional repression by rounding up HP1 proteins to methylated histones. ESET is widely expressed with highest levels found in testis.

Description: Cleavage of the serine proteinase plasminogen to form plasmin is the central event in the dissolution of blood clots by the fibrinolytic system.Plasminogen related protein B is encoded by the PLGLB1 and PLGLB2 genes. It may bind to lysine binding sites present in the kringle structures of plasminogen interfering with the binding of fibrin or alpha-2-antiplasmin to plasminogen. Within the fibrinolytic cascade, the serine proteinases urokinase-type plasminogen activator (uPA) and tissue-type plasminogen activator (tPA) activate the proenzyme plasminogen by cleaving plasminogen to form the fibrinolytically active enzyme plasmin. PLGLB2 (plasminogen-like B2), also known as PLGP1, is a 96 amino acid protein that resembles the N-terminal plasminogen activation peptide, which is released from plasminogen during conversion to plasmin.

Description: Involved in control of the cell cycle. Also antagonizestransactivation by ZBTB17 and GABP2; represses ZBTB17 activation ofthe p15(INK4b) promoter and inhibits its ability to recruit p300.Coactivator for EGR2 and GABP2. Tethers the chromatin modifyingSet1/Ash2 histone H3 'Lys-4' methyltransferase (H3K4me) and Sin3histone deacetylase (HDAC) complexes (involved in the activationand repression of transcription, respectively) together. Componentof a THAP1/THAP3-HCFC1-OGT complex that is required for theregulation of the transcriptional activity of RRM1. As part of theNSL complex it may be involved in acetylation of nucleosomalhistone H4 on several lysine residues. In case of human herpessimplex virus (HSV) infection, HCFC1 forms a multiprotein-DNAcomplex with the viral transactivator protein VP16 and POU2F1thereby enabling the transcription of the viral immediate earlygenes.

Description: Biotin, also known as vitamin B7, is an essential water-soluble vitamin that is a cofactor in glucogenesis and in the metabolism of fatty acids and leucine. Biotinidase is a 523 amino acid enzyme that catalyses the hydrolysis of biocytin to biotin and lysine. Secreted into extracellular space, biotinidase is expressed in liver, heart, placenta, brain, skeletal muscle, pancreas and kidney. Biotinidase contains one carbon-nitrogen hydrolase domain, which is involved in the reduction of organic nitrogen compounds and ammonia production. Defects in the gene encoding biotinidase are the cause of biotinidase deficiency, which is characterised by skin rash, ataxia, seizures, hearing loss, hypotonia and optic atrophy. These symptoms are due to the individual's inability to reutilize biotin and can, therefore, typically be treated with the addition of free biotin.

Description: Polycomb group (PcG) protein. Catalytic subunit of the PRC2/EED-EZH1 complex, which methylates 'Lys-27' of histone H3, leading to transcriptional repression of the affected target gene. Able to mono-, di- and trimethylate 'Lys-27' of histone H3 to form H3K27me1, H3K27me2 and H3K27me3, respectively. Required for embryonic stem cell derivation and self-renewal, suggesting that it is involved in safeguarding embryonic stem cell identity. Compared to EZH1-containing complexes, it is less abundant in embryonic stem cells, has weak methyltransferase activity and plays a less critical role in forming H3K27me3, which is required for embryonic stem cell identity and proper differentiation. Sequence similarities Belongs to the histone-lysine methyltransferase family. EZ subfamily.Contains 1 SET domain.
