Description: The LIM-only (LMO) proteins, LMO1 and LMO2, are nuclear factors that are characterized by a conserved LIM domain. The LIM domain consists of a cysteine-rich zinc-binding motif that is present in a variety of transcription factors, including the LIM homeobox (LHX) proteins expressed in the central nervous system and involved in cell differentiation. LMO1 and LMO2 are expressed in the adult CNS in a cell type-specific manner, where they are differentially regulated by neuronal activity and are involved in regulating the cellular differentiated phenotype of neurons. LMO2 lacks a specific DNA-binding homeobox domain but rather assembles into transcriptional regulatory complexes to mediate gene expression by interacting with the widely expressed nuclear LIM interactor (NLI). NLI, known also as CLIM-1, and the related protein CLIM-2, facilitate the formation of heteromeric LIM complexes and also enhance the nuclear retention of LIM proteins. LMO2 and the related protein LMO4 are expressed in thymic precursor cells. LMO4 is also expressed in mature T cells, cranial neural crest cells, somite, dorsal limb bud mesenchyme, motor neurons, and Schwann cell progenitors.

Description: The calcium-activated chloride channel (CLCA) protein family, which includes the human homologs CLCA1 and CLCA2, display distinct tissue distribution patterns. CLCA1 is expressed as a precursor protein that is processed into two cell surface associated subunits and a group of proteins. CLCA1 is upregulated by interleukin-9 and regulates the expression of mucins. CLCA1 may provide a therapeutic target to control mucus overproduction in airway disease patients with cystic fibrosis. CLCA2 expression is downregulated in breast cancer, therefore CLCA2 is thought to act as a tumor suppressor in normal cells. CLCA3 (known as Clca3 in mouse) is a structurally divergent member of the CLCA family that does not function as a channel protein. CLCA4 is a CLCA member that is expressed in human rectal mucosa, CLCA5 shows strong expression in eye and spleen, and CLCA6 is primarily expressed in intestine and stomach.

Description: Napsin is a pepsin-like aspartic proteinase connected with maturation of surfactant protein B.There are two closely related napsins, napsin A and napsin B. Napsin A is expressed as a single chain protein. Immunohistochemical studies revealed high expression levels of napsin A in human lung and kidney but low expression in spleen. Napsin A is expressed in type II pneumocytes and in adenocarcinomas of lung. The high specificity expression of napsin A in adenocarcinomas of lung is useful to distinguish primary lung adenocarcinomas from adenocarcinomas of other organs.

Description: This MAb recognizes a 21 kDa protein, identified as the p21WAF1 tumor suppressor protein. It is highly specific to p21 and shows no cross-reaction with other closely related mitotic inhibitors. p21WAF1 is a specific inhibitor of cdk s and a tumor suppressor involved in the pathogenesis of a variety of malignancies. The expression of this gene acts as an inhibitor of the cell cycle during G1 phase and is tightly controlled by the tumor suppressor protein p53. Its expression is induced by the wild type, but not mutant, p53 suppressor protein. Normal cells generally display a rather intense nuclear p21 expression. Loss of p21 expression has been reported in many carcinomas (gastric carcinoma, non-small cell lung carcinoma, thyroid carcinoma).

Description: EMP-2 is a 167 amino acid multi-pass membrane protein that contains four-transmembrane domains and belongs to the GAS3/PMP22 (growth arrest-specific-3/peripheral myelin protein-22) family. Localised to lipid raft domains in the plasma membrane, EMP-2 regulates the expression of several target proteins and is necessary for blastocyst implantation in the uterine endometrium. Specifically, EMP-2 mediates blastocyst implantation by controlling the cell membrane expression of MHC and glycosylphosphatidylinositol-anchored proteins, as well as Integrins and caveolin-1. In adult tissues, EMP-2 is expressed in heart, lung, ovary and intestine, while fetal expression is highest in kidney, brain and liver. Overexpression of EMP-2 is associated with endometrial adenocarcinoma, suggesting a possible role for EMP-2 in tumorigenesis.

Description: This gene encodes a tumor suppressor protein that is ubiquitously expressed in normal tissues. In uveal melanomas, expression of this protein is silenced in rapidly metastasizing and metastatic tumor cells but has normal expression in slowly metastasizing or nonmetastasizing tumor cells. This protein may have a role in cell-cycle control by interacting with the Cdk1/cyclinB1 complex. This gene is located on chromosomal region8p22. Loss of heterozygosity (LOH) in the 8p arm is a common characteristic of many types of cancer.

Description: CHRDL2 is a novel chordin like protein that can act as a BMP antagonist. Amember of the chordin family of proteins, it contains a signal peptide andthree CR (cysteine-rich repeat) domains. When expressed as a recombinantprotein it is secreted and binds to activin A, but not to BMP-2, -4, -6. Differential expression has been detected in developing chondrocytes, myoblasts, osteoblasts, and osteoarthritic joints. Complex alternative splicing of CHRDL2 potentially results in distinct isoforms that differ at their C termini, in the expression of signal peptide, and in the content of CR domains. CHRDL2 was originally characterized as a novel protein exclusively expressed in breast, lung, and colon tumors.

Description: Caldesmon, Filamin 1, Nebulin, Plastin, ADF, Gelsolin, CapG, Dematin and Cofilin are differentially expressed Actin-binding proteins. Both muscular (CDh) and non-muscular (CD1) forms of Caldesmon bind to Actin as well as to Calmodulin and Myosin. CDh is expressed predominantly on thin filaments in smooth muscle, whereas CD1 is widely expressed in non-muscle tissues and cells. CapG, also designated Actin-regulatory protein and macrophage-capping protein, is a macrophage-specific protein that reversibly blocks the barbed ends of Actin filaments, but does not sever preformed ones. The interactions of CapG with Actin may be important in the regulation of nuclear and cytoplasmic structures. CapG is a calcium-sensitive DNA-binding protein that plays a role in macrophage function. It is expressed in macrophages and macrophage-like cells and can localize both to the nucleus and the cytoplasm.

Description: KCNF1 is a multi-pass membrane-bound protein that acts as an ion channel and is generally expressed as a heterotetramer of potassium channeling proteins. Formerly known as kH1, KCNF1 is usually found as a heteromer with three other potassium channel proteins, KCNG3, KV6.3 and KCNV2. As a potassium channel protein, KCNF1 plays a role in regulating apoptosis and proliferation of pulmonary artery smooth muscle (PASM) cells. Bone morphogenetic proteins (BMPs) restrict proliferation and can induce apoptosis in normal human PASM cells and will upregulate expression of KCNF1 in PASM cells in vitro. KCNF1 is expressed in heart, brain, liver, skeletal muscle, kidney and pancreas.
