Description: This MAb is specific to c-erbB-2/HER-2 and shows minimal cross-reaction with other members of the family. C-erbB-2/HER-2 is a member of the EGFR family. Receptors of this family are located on the plasma membrane and consist of an extracellular ligand-binding domain that is connected to a large intracellular domain by a single transmembrane sequence. c-erbB-2/HER-2 protein is over-expressed in a variety of carcinomas especially those of breast and ovary.

Description: This antibody recognizes a carbohydrate epitope on a single chain, transmembrane, heavily glycosylated protein of 90-120 kDa, which is identified as CD34 (VI international workshop on human differentiation antigens). Its expression is a hallmark for identifying pluripotent hematopoietic stem or progenitor cells. Its expression is gradually lost as lineage committed progenitors differentiate. CD34 is a marker of choice for staining blasts in acute myeloid leukemia. In addition, it is expressed by soft tissue tumors, such as solitary fibrous tumor and gastrointestinal stromal tumor. CD34 expression is also found in vascular endothelium. Additionally, proliferating endothelial cells overexpress this molecule than the non-proliferating endothelial cells. Anti-CD34 labels > 85% of angiosarcoma and Kaposi's sarcoma, but shows low specificity.

Description: The highly leukemogenic avian retrovirus E26 contains two oncogenes, v-Myb and v-Ets, which are expressed together as a fusion protein. The cellular homolog of v-Myb, designated c-Myb, encodes a transcription factor. Deletion or disruption of a negative regulatory domain mapping within the carboxy-terminal domain of c-Myb results in enhanced transactivating capacity and in parallel, leads to activation of its ability to transform hemopoietic cells. c-Myb is expressed preferentially, but not exclusively, in immature hemopoietic cells and its expression decreases as cells differentiate.

Description: Multiple isoelectric variants of calponin have been identified, however only two molecular weight isoforms exist; a 34 kDa form and a 29 kDa form. Expression of the 29 kDa form, I-calponin, is primarily restricted to muscle of the urogenital tract, whereas the higher molecular weight variant has been demonstrated in vascular and visceral smooth muscle. In Western blotting, this MAb reacts with only the 34 kDa form of calponin in extracts of human aortic medial smooth muscle and is unreactive with fibroblast extracts of cultivated human foreskin. Calponin is a calmodulin, F-actin and tropomyosin binding protein, which is thought to be involved in the regulation of smooth muscle contraction. Calponin expression is restricted to smooth muscle cells and has been shown to be a marker of the differentiated (contractile) phenotype of developing smooth muscle.

Description: This antibody recognizes a glycoprotein of 110 kDa, which is identified as CD68. It is important for identifying macrophages in tissue sections. It stains macrophages in a wide variety of human tissues, including Kupffer cells and macrophages in the red pulp of the spleen, in lamina propria of the gut, in lung alveoli, and in bone marrow. It reacts with myeloid precursors and peripheral blood granulocytes. It also reacts with plasmacytoid T cells, which are supposed to be of monocyte/macrophage origin. It shows strong granular cytoplasmic staining of chronic and acute myeloid leukemia and also reacts with rare cases of true histiocytic neoplasia. Lymphomas are negative or show few granules.

Description: This antibody recognizes CD16 (FcγRIII), the low-affinity receptor for IgG with an apparent molecular weight of 50-80 kDa. Two similar genes represent CD16, CD16A (FcγRIIIA), which exists as a hetero-oligomeric polypeptide-anchored form in macrophages and NK cells and CD16B (FcγRIIIB), which exist as a monomeric GPI-anchored form in neutrophils. Furthermore, there are two known polymorphisms of CD16B, NA-1 and NA-2. Individuals homozygous for NA-2 show a lower phagocytic capacity compared with NA-1. CD16 binds IgG in the form of immune complexes and shows preferential binding of IgG1 and IgG3 isotypes and minimal binding of IgG2 and IgG4. Upon IgG binding, both CD16 isoforms initiate signal transduction cascades that lead to a variety of responses including antibody-dependent cell-mediated cytotoxicity (ADCC), phagocytosis, degranulation and proliferation.

Description: This antibody recognizes CK1, CK5, CK10 and CK14. In normal epithelia, it stains stratified epithelia, myoepithelial cells and basal cells in the prostate gland and bronchi. This MAb shows no reactivity with hepatocytes, pancreatic acinar cells, proximal renal tubules, or endometrial glands; there is no reactivity with cells derived from simple epithelia. Mesenchymal tumors, lymphomas, melanomas, neural tumors, and neuroendocrine tumors are negative with this antibody. It stains myoepithelial cells and has been shown to be useful in distinguishing prostate adenocarcinoma from benign prostate. This antibody has also been useful in separating benign from malignant intraductal breast proliferations.

Description: Recognizes the L1 or Calprotectin molecule, an intra-cytoplasmic antigen comprising of a 12 kDa alpha chain and a 14 kDa beta chain expressed by granulocytes, monocytes and by tissue macrophages. Macrophages usually arise from hematopoietic stem cells in the bone marrow. Under migration into tissues, the monocytes undergo further differentiation to become multifunctional tissue macrophages. They are classified into normal and inflammatory macrophages. Normal macrophages include macrophages in connective tissue (histiocytes), liver (Kupffer's cells), lung (alveolar macrophages), lymph nodes (free and fixed macrophages), spleen (free and fixed macrophages), bone marrow (fixed macrophages), serous fluids (pleural and peritoneal macrophages), skin (histiocytes, Langerhans's cell) and in other tissues. Inflammatory macrophages are present in various exudates. Macrophages are part of the innate immune system, recognizing, engulfing and destroying many potential pathogens including bacteria, pathogenic protozoa, fungi and helminthes. This MAb reacts with neutrophils, monocytes, macrophages, and squamous mucosal epithelia and has been shown as an important marker for identifying macrophages in tissue sections.

Specificity of this MAb is validated by sequential immunoprecipitation with a PAb against Tenascin C. Tenascin C is a multifunctional, disulfide-linkedhexameric extracellular matrix glycoprotein expressed in association with mesenchymal epithelial interactions during development and in the neo-vasculature and stroma of undifferentiated tumors. In adults, it is restricted to certain epithelial-stromal interfaces and increases markedly in hyper-proliferative diseases and in stroma of many neoplasms, including gliomas, breast, squamous and lung carcinomas.

Description: Epstein-Barr virus (EBV), also designated human herpesvirus 4 (HHV-4), is a member of the herpesvirus family and is one of the most common human viruses. EBV infects B cells and, though often asymptomatic, it can cause infectious mononucleosis, a disease characterized by fatigue, fever, sore throat and muscle soreness. The EBV-induced early antigens (Ea) are among several antigen complexes that have been identified in EBV-infected cells. The Ea complex is composed of diffuse (Ea-D) and restricted (Ea-R) components. The activity of Ea-D is suppressed during latent infection. BMRF1, the gene that encodes for Ea-D, is closely associated with the gene encoding for EBV DNA polymerase, and Ea-D is essential for the activity of this polymerase. Ea-D forms a complex with EBV DNase and, together, they may play a role in viral replication.

Description: MAb IPO-10 defines the antigen, which appears on B cell progenitors following HLA-DR and preceding CD10, CD19, CD22, CD37 and cym. It is expressed on resting B cells and than reappears and persists in cytoplasm and on cell surface until cytoplasmic Ig appears. It is a useful antibody for diagnostics of neoplasms of B cell origins. It reacts with human B cell lines Daudi, Raji, Namalva, EB-3, RPMI-8226 (50% of cells). The MAb does not label T cell lines, blood granulocytes, thymocytes or bone marrow stromal fibroblasts. No significant changes are detected after PHA or ConA stimulation while LPS and PWM stimulated cultures after 18-48h show decreased number of antigen-positive cells but in final terms of cultivation antigen is expressed again. This MAb labels B cell leukemias and some lymphomas. Hairy cell leukemia strongly reacts and 70% of B cell CLL and some B-NHL were also positive. IPO-10 reacts with AMML cells and in a majority of Hodgkin's disease cases a significant percentage of affected lymph node cells were detected.

Description: Recognizes a protein of 104 kDa-110 kDa, characterized as major vault protein (MVP). Vaults are large ribonucleoprotein particles (RNPs) present in all eukaryotic cells. They have a complex morphology, including several small molecules of RNA, but a single protein species. The MVP accounts for >70% of their mass. Their shape is reminiscent of the nucleopore central plug. Treatment of cells with estradiol increases the amount of MVP in nuclear extract. The hormone-dependent interaction of vaults with ER is prevented in vitro by sodium molybdate. Antibodies to estrogen, progesterone and glucocorticoid receptors are able to co-immunoprecipitate the MVP. MVP is overexpressed in many neoplastic tissues and cell lines. Expression of MVP predicts a poor response to chemotherapy.

Lutropin (also designated luteinizing hormone) plays a role in spermatogenesis and ovulation by stimulating the testes and ovaries to produce steroids. Gonadotropin (also designated choriogonadotropin) production in the placenta maintains estrogen and progesterone levels during the first trimester of pregnancy. Ovaries and testes abundantly express luteinizing hormone/choriogonadotropin receptor. GnRH receptor contains seven hydrophobic transmembrane domains connected by hydrophilic extracellular and intracellular loops characteristic of G-protein coupled receptors. GnRH stimulates the gonadotrophs of the anterior pituitary to secrete luteinizing hormone (LH) as well as follicle-stimulating hormone (FSH). GnRH influences the protective effect of pregnancy and Gonadotropin against breast cancer. The expression of GnRH on breast carcinoma correlates in part to the degree of tumor differentiation. GnRH-positive breast tumors occur more frequently in tumors with greater cell differentiation in premenopausal women. GnRH is present in luteal and granulosa cells as well as in ovarian cell membrane preparations.
