Description: MAb VU-2G7 reacts with MUC1, a large transmembrane glycoprotein expressed on the ductal surface of normal glandular epithelia. The dominant epitope of VU-2G7 has not been established with epitope fingerprinting. The extracellular domain of MUC1 largely consists of a highly conserved, O-glycosylated 20 amino acids tandem repeat which can occur 30-100 times per molecule depending on the length of the allele involved. In the vast majority of human carcinomas this protein is up regulated and poorly glycosylated and appears on the cell surface in a non-polarized fashion.

Description: Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI 6.0) subfamilies. This antibody recognizes acidic (Type I or LMW) and basic (Type II or HMW) cytokeratins, including 59 kDa (CK4); 58 kDa (CK5); 56 kDa (CK6); 52 kDa (CK8); 56.5 kDa (CK10); 53 kDa (CK13) and 45 kDa (CK18). This is a broad-spectrum antibody, which has been reported to differentiate epithelial tumors from non-epithelial tumors. Many studies have shown the usefulness of keratins as markers in cancer research and tumor diagnosis.

Description: This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. Expression of CK10 is related to maturation of malignant keratinocytes, being preferentially detected in more-differentiated parts.

Description: This antibody reacts with Ig domain of CD47 protein. It has been shown to inhibit polymorphonuclear neutrophil (PMN) transmigration across cell monolayers and matrix. CD47, originally named integrin-associated protein (IAP), is a 50 kDa protein containing five membrane-spanning sequences and a short cytoplasmic tail. CD47 plays a role in both cell adhesion by acting as an adhesion receptor for THBS1 on platelets, and in the modulation of integrins. It is important in memory formation and synaptic plasticity in the hippocampus. CD47 may play a role in membrane transport and/or integrin dependent signal transduction.

Description: Recognizes a protein of 75 kDa, identified as mu heavy chain of human immunoglobulins. It does not cross-react with alpha (IgA), gamma (IgG), epsilon (IgE), or delta (IgD), heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. This MAb is useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. The most common feature of these malignancies is the restricted expression of a single heavy chain class. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is clonal and therefore malignant.

Description: Recognizes a protein of 210-220 kDa, which is identified as the complement receptor 1 (CR1)/CD35. This MAb does not block CR1 activity. It is highly specific to CR1 and shows no cross-reaction with CR2. The primary function of CR1 is to serve as the cellular receptor for C3b and C4b, the most important components of the complement system leading to clearance of foreign macromolecules. The Knops blood group system is a system of antigens located on this protein. Follicular dendritic cells (FDC) are restricted to the B-cell regions of secondary lymphoid follicles. They are CD21 /CD35 /CD1a-. This MAb labels follicular dendritic cells and follicular dendritic cell sarcoma.

Description: ZAP70 is a 70 kDa protein tyrosine kinase found in T-cells and natural killer cells. Control of this protein translation is via the IgVH gene.In Western blotting of whole cell lysates of normal peripheral blood mononuclear cells, the antibody labels a band corresponding to ZAP70. In Western blotting of whole cell lysates of CD19-positive purified leukemia cells from patients with Ig-unmutated and Ig-mutated CLL, the antibody labels a band corresponding to ZAP70 in the Ig-unmutated CLL samples, whereas no band is observed in the Ig-mutated CLL samples. In Western blotting of cell lysates of Jurkat cells (T-lymphoblastic cell line), the antibody labels a band of 70 kDa protein. In Western blotting of cell lysates of A431 cells (carcinoma cell line), no band is observed. ZAP70 protein is expressed in leukemic cells of approximately 25% of chronic lymphocytic leukemia (CLL) cases as well. Anti-ZAP70 expression is an excellent surrogate marker for the distinction between the Ig-mutated (anti-ZAP70 negative) and Ig-unmutated (anti-ZAP70 positive) CLL subtypes and can identify patient groups with divergent clinical courses. The anti-ZAP70 positive Ig-unmutated CLL cases have been shown to have a poorer prognosis.

Description: This antibody recognizes cytokeratin 10 (56.5 kDa) and cytokeratin 13 (53 kDa) in Western blotting. It recognizes only cytokeratin 13 in formalin-fixed, paraffin-embedded tissue sections. It does not react with cytokeratin 10 positive, cytokeratin 13 negative epithelia such as epidermis. However, on frozen sections this MAb serves as differentiation-related marker of all stratified epithelia; it stains all suprabasal cells in both cornifying and non-cornifying stratified epithelia and more differentiated cells of squamous carcinomas.

It is expressed on platelets, monocytes and macrophages, microvascular endothelial cells, erythrocyte precursors, mammary epithelial cells, and some macrophage derived dendritic cells. CD36 acts as a receptor for thrombospondin (TSP), collagen types I, IV and V, P. falciparum malaria-infected erythrocytes, and sickle erythrocytes. It also functions as a scavenger receptor, mediating macrophage uptake of oxidized low-density lipoprotein (LDL) and recognition of apoptotic polymorphonuclear leukocytes (PMN). CD36 plays a role in platelet aggregation, macrophage foam cell development, inflammation, and the tissue ischemia observed in sickle cell disease and cerebral malaria. Note that 1-4% of Japanese and East Asia population lack CD36.
