Description: In Western blotting, this antibody detects an antigen of 125 kDa in human liver and 135 kDa in tumors of histiocytic origin. Comparative study of this MAb and a standard CD68 MAb showed that their antigens are different. Its antigen in all macrophage types studied is located on the plasma membrane and within cytoplasmic structures including lysosomes. This MAb shows a restricted reactivity to cells of the monocyte/macrophage system. It specifically reacts with blood monocytes and stains resident macrophages in a wide variety of human tissues. This MAb does not stain antigen-presenting cells, e.g., Langerhans cells. Reportedly, its reactivity is restricted to histiocytes and macrophages.

Description: Hoechst dyes are cell membrane-permeant, minor groove binding blue fluorescent DNA stains. Hoechst 33258 and Hoechst 33342 are spectrally similar. Hoechst 33528 is slightly more water soluble than Hoechst 33342, but both dyes are widely used in cell cycle and apoptosis studies and also as nuclear counterstains. They are typically used to stain live or fixed cells in buffer or medium at 1 ug/mL, with no wash step required.

Description: This antibody reacts with a 75 kDa melanocyte-specific gene product, identified as Tyrosinase-related protein-1 (TRP-1). It is involved in melanin synthesis. TRP1 is present on the melanosomal membranes of melanoma, normal melanocytes and nevi.Recent evidence suggests that TRP-1 is involved in maintaining stability of tyrosinase protein and modulating its catalytic activity. TRP-1 is also involved in maintenance of melanosome ultrastructure and affects melanocyte proliferation and cell death.

Description: Recognizes a protein of 110 kDa, which is identified as androgen receptor (AR). It reacts with full length, and the newly described A form of the receptor. It does not cross react with estrogen, progesterone, or glucocorticoid receptors. The expression of AR is reportedly inversely correlated with histologic grade i.e. well differentiated prostate tumors show higher expression than the poorly differentiated tumors. In prostate cancer, AR has been proposed, as a marker of hormone-responsiveness and thus it may be useful in identifying patients likely to benefit from anti-androgen therapy. A high molecular weight heterodimeric protein, it was originally discovered as a protein missing from the surfaces of virus-transformed cells, and it has been shown to be involved in various functions including cell adhesion, cell motility and wound healing. Alternative splicing and glycosylation give rise to several different forms of Fibronectin, some of which exhibit restricted tissue distribution or association with malignancies. It has been shown that Myofibroblasts phenotype formation correlates with the occurrence of glycosylated Fibronectin and Fibronectin splice variants in Dupuytren's disease.

Description: Recognizes a protein of 32 kDa, identified as CD8a (also known as CD8 chain, T cell co-receptor, Leu2, and T8). CD8 molecule consists of two chains, termed and chain, which are expressed as a disulphide-linked heterodimer or as an homodimer. CD8 is expressed on T cell subset (cytotoxic/suppressor T cells), thymocytes and NK cells. The majority of CD8 T-cells expresses CD8 as heterodimer. Some subpopulation of CD8 T cells as well as NK cells may express homodimer. CD8 functions as a co-receptor in concert with TCR for binding the MHC class I/peptide complex. The HIV-2 envelope glycoprotein binds CD8 chain (but not chain). The cytoplasmic domain of CD8 associates with p56lck tyrosine kinase.

Description: Recognizes proteins of 33, 35 and 41 kDa, which are identified as various isoforms of CD74. Its epitope is localized in the extracellular domain of CD74. CD74 is a type II transmembrane protein which binds to the peptide binding groove of newly synthesized MHC class II alpha/beta heterodimers and prevents their premature association with endogenous polypeptides. The CD74 molecule plays a critical role in the presentation of peptides, by the MHC class II antigens, to CD4 positive lymphocytes. CD74 is expressed on MHC class II positive cells including B cells, a subset of activated T cells, monocytes, and dendritic cells and by various types of carcinomas. CD74 is expressed primarily by antigen presenting cells, such as B-lymphocytes (from before the pre-B cell stage to before the plasma cell stage), macrophages, and monocytes, and many epithelial cells. Anti-CD74 stains predominantly germinal center lymphocytes and B-cell lymphomas, but rarely T-cell lymphomas. Anti-CD74 has been shown to be useful in differentiating atypical fibroxanthoma (-) from malignant fibrous histiocytoma ( ).

Description: Hepatocyte Specific Antigen, also called Hepatocyte Paraffin 1 or HepPar1, localizes to the mitochondria of hepatocytes. It is a sensitive marker for distinguishing hepatocellular carcinomas (HCC) from other metastatic carcinomas as well as cholangio-carcinomas. HCC's occur primarily in the stomach, but they are also found in many other organs. The Hepatocyte Specific Antigen may also be a useful marker for intestinal metaplasia. Reportedly, strong expression of the Hepatocyte Specific Antigen correlates with smaller tumor size and longer patient survival. Occasionally, Hepatocyte Specific Antigen is also found in gastric carcinomas as well as in a few other non-hepatic tumors.

Description: Recognizes an oncofetal antigen of 220 kDa, identified as a tumor-associated glycoprotein (TAG-72) with properties of a mucin. The majority of human adenocarcinomas including colorectal, pancreatic, gastric, ovarian, endometrial, mammary, and non-small cell lung cancer display some cell populations that are positive for TAG72. About 60% of carcinoma patients express TAG72 in their sera. TAG72 is reportedly useful in distinguishing pulmonary adenocarcinomas from pleural mesotheliomas.

Description: This antibody recognizes a protein of 66 kDa-170 kDa, identified as involucrin. In Western blotting of cultured human keratinocytes, this MAb reacts with a 120 kDa protein. Involucrin is expressed in a range of stratified squamous epithelia, including the cornea, which lacks a distinct cornified layer. In normal epidermis, it is first expressed in the upper spinous layers, and in keratinocyte cultures, all cells that have left the basal layer express it. Involucrin expression is altered in pathological conditions: in psoriasis and other benign epidermal hyperplasias, involucrin expression begins closer to the basal layer than normal; expression is abnormal in squamous cell carcinomas and premalignant lesions, and is reduced in severe dysplasias of the larynx and cervix.

Description: This MAb reacts with an N-terminal epitope (aa 16-25) of both wild type and mutated p53. Mutation and/or allelic loss of p53 is one of the causes of a variety of mesenchymal and epithelial tumors. If it occurs in the germ line, such tumors run in families. In most transformed and tumor cells the concentration of p53 is increased 51000 fold over the minute concentrations (1000 molecules cell) in normal cells, principally due to the increased half-life (4 h) compared to that of the wild-type (20 min). p53 Localizes in the nucleus, but is detectable at the plasma membrane during mitosis and when certain mutations modulate cytoplasmic/nuclear distribution. Mutations arise with an average frequency of 70% but incidence varies from zero in carcinoid lung tumors to 97% in primary melanomas. High concentrations of p53 protein are transiently expressed in human epidermis and superficial dermal fibroblasts following mild ultraviolet irradiation. Positive nuclear staining with p53 antibody has been reported to be a negative prognostic factor in breast carcinoma, lung carcinoma, colorectal, and urothelial carcinoma. Anti-p53 positivity has also been used to differentiate uterine serous carcinoma from endometrioid carcinoma as well as to detect intratubular germ cell neoplasia.
