Description: The epitope of this MAb maps in the C-terminus of PTH, a hormone produced by the parathyroid gland that regulates the concentration of calcium and phosphorus in extracellular fluid. This hormone elevates blood Ca2 levels by dissolving the salts in bone and preventing their renal excretion.It is produced in the parathyroid gland as an 84 amino acid single chain polypeptide. It can also be secreted as N-terminal truncated fragments or C-terminal fragments after intracellular degradation, as in case of hypercalcemia. Defects in this gene are a cause of familial isolated hypoparathyroidism (FIH); also called autosomal dominant hypoparathyroidism or autosomal dominant hypocalcemia. FIH is characterized by hypocalcemia and hyperphosphatemia due to inadequate secretion of parathyroid hormone. Symptoms are seizures, tetany and cramps. FIH exist both as autosomal dominant and recessive forms of hypoparathyroidism.

Description: Recognizes an oncofetal antigen of 220 kDa, identified as a tumor-associated glycoprotein (TAG-72) with properties of a mucin. This MAb defines the mucin-carried sialylated-Tn epitope. TAG-72 is usually expressed by adenocarcinomas, but is negative in mesotheliomas. Studies have reported that this antibody has 80% sensitivity and 93% specificity for pulmonary adenocarcinoma. Therefore, TAG-72 is a useful marker to distinguish between mesothelioma and adenocarcinoma. However, false positive reactions can occur so results must be interpreted with the utmost caution. This antibody may be useful in the differentiation of non-small cell carcinomas from small cell carcinomas of the lung. The combined use of anti-TAG-72 and anti-GCDFP-15 is valuable in the diagnosis of apocrine carcinoma.

Description: Actin is a major component of the cytoskeleton and is present in most cell types. It is highly specific to actin from smooth muscles. This MAb does not stain cardiac or skeletal muscle; however, it does stain myofibroblasts and myoepithelial cells. This antibody could be used together with anti-muscle specific actin and myogenin in making a diagnosis of smooth muscle and skeletal muscle tumors. In most cases of rhabdomyosarcoma, this antibody yields negative results whereas anti-muscle specific actin and myogenin are positive. Leiomyosarcomas are positive only with anti-muscle specific actin and anti-smooth muscle actin and are negative with anti-myogenin.

Description: This antibody recognizes a protein of 66 kDa-170 kDa, identified as involucrin. In Western blotting of cultured human keratinocytes, this MAb reacts with a 120 kDa protein. It stains the involucrin in a variety of sizes: 170 kDa in MCF-7 cells, a doublet of ~115 kDa and 150 kDa in gorilla and owl monkey, 66 kDa in dog, and a doublet of 105 kDa in pig. Its epitope maps between codon 421-568 of human involucrin. Involucrin is expressed in a range of stratified squamous epithelia, including the cornea, which lacks a distinct cornified layer. In normal epidermis, it is first expressed in the upper spinous layers, and in keratinocyte cultures, all cells that have left the basal layer express it. Involucrin expression is altered in pathological conditions: in psoriasis and other benign epidermal hyperplasias, involucrin expression begins closer to the basal layer than normal; expression is abnormal in squamous cell carcinomas and premalignant lesions, and is reduced in severe dysplasias of the larynx and cervix.

Description: This antibody recognizes a protein of 35 kDa, which is identified as tartrate-resistant acid phosphatase (TRAcP). It exists as two isoforms (5a and 5b). This MAb reacts with both the isoforms. Serum TRAcP 5a is secreted by macrophages and dendritic cells and increased in many patients of rheumatoid arthritis.Serum TRAcP 5b is produced from osteoclasts and elevated during bone resorption. TRAcP is an iron containing glycoprotein, which catalyzes the conversion of orthophosphoric monoester to alcohol and orthophosphate. It is the most basic of the acid phosphatases and is the only form not inhibited by L( )-tartrate. TRAcP is synthesized as a latent proenzyme and is activated by proteolytic cleavage and reduction. Normally, TRAcP is highly expressed by osteoclasts, activated macrophages, neurons and endometrium during pregnancy. Expression of TRAcP is increased in certain pathological conditions such as Leukemic Reticuloendotheliosis (Hairy Cell Leukemia), Gaucher's Disease, HIV-induced Encephalopathy, Osteoclastoma and in osteoporosis and metabolic bone diseases. Anti-TRAcP antibody labels the cells of Hairy Cell Leukemia (HCL) with a high degree of sensitivity and specificity. Other cells stained with this antibody are tissue macrophages and osteoclasts.

Description: Recognizes a protein of 110 kDa, which is identified as androgen receptor (AR). It reacts with full length, and the newly described A form of the receptor. It does not cross react with estrogen, progesterone, or glucocorticoid receptors. The expression of AR is reportedly inversely correlated with histologic grade i.e. well differentiated prostate tumors show higher expression than the poorly differentiated tumors. Description: This MAb reacts with a wide variety of simple epithelia. It does not react with stratified squamous epithelia. It reacts with epithelial tumors of the gastrointestinal tract, lung, breast, pancreas, ovary, and thyroid. Cytokeratin 18, which belongs to the type A (acidic) subfamily of low molecular weight keratins, exists in combination with cytokeratin 8. It is reported that tissues from gastrointestinal tract are positive for both cytokeratin 8 and 18 but do not contain cytokeratin 14. Tissues from gastrointestinal tract, respiratory tract and urogenital tract, as well as endocrine and exocrine tissues and mesothelial cells are positive for cytokeratin 18.

Description: Recognizes a 47-55 kDa-tumor suppressor protein, identified as Wilm's Tumor (WT1) protein. The antibody reacts with all isoforms of the full-length WT1 and also identifies WT1 lacking exon 2-encoded amino acids, frequently found in subsets of sporadic Wilm s tumors.WT1, a sporadic and familial pediatric kidney tumor, is genetically heterogeneous. Wilm s tumor is associated with mutations of WT1, a zinc-finger transcription factor that is essential for the development of the metanephric kidney and the urogenital system. The WT1 gene is normally expressed in fetal kidney and mesothelium, and its expression has been suggested as a marker for Wilm s tumor and mesothelioma. WT1 protein has been identified in proliferative mesothelial cells, malignant mesothelioma, ovarian carcinoma, gonadoblastoma, nephroblastoma, and desmoplastic small round cell tumor. Lung adenocarcinomas rarely stain positive with this antibody. WT1 protein expression in mesothelial cells has become a reliable marker for the diagnosis of mesotheliomas.

This MAb is specific to Synacthen (aa1-24 of ACTH); does not react with CLIP (aa17-39 of ACTH). POMC (pro-opiomelanocortin or corticotropin-lipotropin) is a 267 amino acid polypeptide hormone precursor that goes through extensive, tissue-specific posttranslational processing by convertases. POMC is cleaved into ten hormone chains named NPP, ACTH, alpha-MSH (Melanocyte Stimulating Hormone), beta-MSH, gamma-MSH, CLIP (corticotropin-like intermediary peptide), Lipotropin-beta, Lipotropin-gamma, beta-endorphin and Met-enkephalin. ACTH is also produced by cells of immune system (T-cells, B-cells, and macrophages) in response to stimuli associated with stress. Anti-ACTH is a useful marker in classification of pituitary tumors and the study of pituitary disease. It reacts with ACTH-producing cells (corticotrophs).It also may react with other tumors (e.g. some small cell carcinomas of the lung) causing paraneoplastic syndromes by secreting ACTH.
