Description: This MAb recognizes a 60 kDa antigen associated with the mitochondria in human cells. It is a part of a new panel of reagents, which recognizes subcellular organelles or compartments of human cells. These markers may be useful in identification of these organelles in cells, tissues, and biochemical preparations. It recognizes an antigen associated with the mitochondria in human cells only. It can be used to stain the mitochondria in cell or tissue preparations and can be used as a mitochondrial marker in subcellular fractions. It produces a spaghetti-like pattern in normal and malignant cells and may be used to stain mitochondria of cells in fixed or frozen tissue sections. It can also be used with paraformaldehyde fixed frozen tissue or cell preparations. This MAb is an excellent marker for human cells in xenographic model research. It reacts specifically with human cells, including neurons and embryonic stem cells.

Description: This MAb recognizes a single chain, transmembrane, heavily glycosylated protein of 90-120 kDa, which is identified as CD34. Its expression is a hallmark for identifying pluripotent hematopoietic stem or progenitor cells. Its expression is gradually lost as lineage committed progenitors differentiate. CD34 is a marker of choice for staining blasts in acute myeloid leukemia. In addition, CD34 is expressed by soft tissue tumors, such as solitary fibrous tumor and gastrointestinal stromal tumor. Its expression is also found in vascular endothelium. Additionally, it appears that proliferating endothelial cells express this molecule more than the non-proliferating endothelial cells. Anti-CD34 labels > 85% of angiosarcoma and Kaposi's sarcoma, but with a lower specificity.

Description: This antibody recognizes a cell surface glycoprotein of 95/115/135 kDa (depending upon the extent of glycosylation), identified as CD43 [Workshop IV]. Epitope of MAb Bra7G is clearly different from that of MAb DF-T1, called b as opposed to a for DF-T1. 70-90% of T-cell lymphomas and from 22-37% of B-cell lymphomas express CD43. No reactivity has been observed with reactive B-cells. So a B-lineage population that co-expresses CD43 is highly likely to be a malignant lymphoma, especially a low-grade lymphoma, rather than a reactive B-cell population. When CD43 antibody is used in combination with anti-CD20, effective immunophenotyping of the lymphomas in formalin-fixed tissues can be obtained. Co-staining of a lymphoid infiltrate with anti-CD20 and anti-CD43 argues against a reactive process and favors a diagnosis of lymphoma.

Description: Recognizes a protein of 150 kDa, which is identified as the high molecular weight variant of Caldesmon. Two closely related variants of human caldesmon have been identified which are different in their electrophoretic mobility and cellular distribution. The h-caldesmon variant (120-150 kDa) is predominantly expressed in smooth muscle whereas l-caldesmon (70-80 kDa) is found in non- muscle tissue and cells. Neither of the two variants has been detected in skeletal muscle. This MAb recognizes only the 150 kDa variant (h-caldesmon) in Western blots of human aortic media extracts and is unreactive with fibroblast extracts from cultivated human foreskin. Caldesmon is a developmentally regulated protein involved in smooth muscle and non-muscle contraction.

Description: Recognizes a cell surface glycoprotein of 80-95 kDa (CD44) on lymphocytes, monocytes, and granulocytes (Leucocyte Typing Workshop V). Its epitope is resistant to digestion by trypsin and chymotrypsin. This MAb selectively interferes with lymphocyte binding to lymph node, mucosal and synovial endothelium. The CD44 family of glycoproteins exists in a number of variant isoforms, the most common being the standard 85-95 kDa or hematopoietic variant (CD44s). Higher molecular weight isoforms are described in epithelial cells (CD44v), which are believed to function in intercellular adhesion and stromal binding. CD44 immunostaining is commonly used for the discrimination of urothelial transitional cell carcinoma in-situ from non-neoplastic changes in the urothelium.

Description: Recognizes a 53 kDa protein, which is identified as p53 suppressor gene product. It reacts with the mutant as well as the wild form of p53 under denaturing and non-denaturing conditions. p53 is a tumor suppressor gene expressed in a wide variety of tissue types and is involved in regulating cell growth, replication, and apoptosis. It binds to MDM2, SV40 T antigen and human papilloma virus E6 protein. Positive nuclear staining with p53 antibody has been reported to be a negative prognostic factor in breast carcinoma, lung carcinoma, colorectal, and urothelial carcinoma. Description: This MAb reacts with a 28 kDa chain of HLA-DRB1 antigen, a member of MHC class II molecules. It does not cross react with HLA-DP and HLA-DQ. The L243 antibody recognizes a different epitope than the LN3 monoclonal antibody, and these antibodies do not cross-block binding to each other's respective epitopes. HLA-DR is a heterodimeric cell surface glycoprotein comprised of a 36 kDa alpha (heavy) chain and a 28 kDa beta (light) chain. It is expressed on B-cells, activated T-cells, monocytes/macrophages, dendritic cells and other non-professional APCs. In conjunction with the CD3/TCR complex and CD4 molecules, HLA-DR is critical for efficient peptide presentation to CD4 T cells. It is an excellent histiocytic marker in paraffin sections producing intense staining. True histiocytic neoplasms are similarly positive. HLA-DR antigens also occur on a variety of epithelial cells and their corresponding neoplastic counterparts. Loss of HLA-DR expression is related to tumor microenvironment and predicts adverse outcome in diffuse large B-cell lymphoma.

Description: This antibody recognizes a 56 kDa protein, which is identified as Early Mitotic Inhibitor-1 (EMI1). It regulates mitosis by inhibiting the anaphase promoting complex/cyclosome (APC). Emi1 is a conserved F box protein containing a zinc-binding region essential for APC inhibition. The Emi1 protein functions to promote cyclin A accumulation and S phase entry in somatic cells by inhibiting the APC complex. At the G1-S transition, Emi1 is transcriptionally induced by the E2F transcription factor. Emi1 overexpression accelerates S-phase entry and can override a G1 block caused by overexpression of Cdh1 or the E2F-inhibitor p105 retinoblastoma protein (pRb). Depleting cells of Emi1 through RNA interference prevents accumulation of cyclin A and inhibits S phase entry.

Description: Recognizes a 180-185 kDa protein, identified as isoform of leukocyte common antigen (CD45RO) (4th Leucocyte Typing Workshop: Code No. N31). The epitope recognized by this antibody is sensitive to neuraminidase digestion. This antibody reacts with mature activated T-cells, most thymocytes, and a sub-population of resting T-cells within both CD4 and CD8 subsets. It shows no reactivity with normal B or natural killer cells, but reacts with granulocytes and monocytes. Reportedly, it is useful to identify T-cell lymphomas and leukemias. It rarely stains NK cells or B-cell lymphomas.

Description: BCL10, with an N-terminal caspase recruitment domain (CARD), is found in a number of apoptotic regulatory molecules. It was identified through its direct involvement in t(1;14) of mucosa-associated lymphoid tissue (MALT) lymphoma. Expression of BCL10 was shown to induce NFkB activation in a NIK-dependent pathway. This MAb labels subpopulations of normal B and T cells and is a useful tool for the sub-classification of lymphomas. In MALT lymphomas with the t(1;14) translocation, while 55% of MALT lymphomas lacking this translocation exhibited the same labeling pattern, although at a much lower level.

Description: Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Most pituitary adenomas and prolactinomas readily express chromogranin.

TS converts deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP), which is essential for DNA biosynthesis. TS is also a critical target for the fluoropyrimidines, an important group of antineoplastic drugs that are widely used in the treatment of solid tumors. Both 5-FU and fluorodeoxyuridine are converted in tumor cells to FdUMP which inactivates TS by formation of a ternary covalent complex in the presence of the folate cofactor 5,10-methylenetetrahydrofolate. Expression of TS protein is associated with response to 5-fluorouracil (5-FU) in human colorectal, gastric, head and neck, and breast carcinomas.
